I don't recommend using touchscreen controls. You get less tactile feedback, you cover part of the screen, it's harder to hold the system, etc. If you must, you should use as Kolma described (use left hand for lower and left enemies, right hand for upper and right, or something like that) or hold in one hand while using multiple fingers on your other hand.
The way I do recommend playing is using both sets of buttons and using alternating thumbs to hit enemies (and then using both simultaneously on red enemies).
I'm not sure that having button icons on the sides of the screen to touch would work out well for most people, since they are less intuitive in terms of representing enemy direction.

When i try using the Touchscreen to tap out the beats it doesn't feel as intuitive. I can reach the Square and Circle prompts fine with my thumbs but X and Triangle are a bit trickier.
