i think the best way is to get it on survival, the combo continues between songs if i remember right.

For me. Pendulum - Propane Nightmares ( Celldweller rmx) was the level, where I got the 500 hit combo. Play on expert, and you have about 550 enemies to hit.

Right now, I'm trying to get the perfect hits on Shen Yi - War Dance, but there is a twist. You don't have to get all the orbs, since they just decrease your combo, and not counted as fail
